Direct answer (40-60 words)
Yes. Novo Nordisk's official Wegovy instructions allow priming up to three times if no drop of medication appears at the needle tip. Each priming attempt should use a fresh needle and the flow-check symbol on the dose dial. If three primes produce no drop, discard the pen and contact the pharmacy for a replacement.

What priming actually does
A Wegovy pen is a pre-filled, single-patient injectable device. Each pen contains the medication, a plunger mechanism, and a dose-selection dial. Before the first injection from a pen, you have to confirm that the medication is flowing correctly through the device.
Check your GLP-1 eligibility
Use our free BMI Calculator to see if you may qualify for provider-reviewed GLP-1 therapy.
Try the BMI Calculator →Priming serves two functions:
- Removes air from the needle and cartridge channel. Air pockets can collect at the top of the cartridge during shipping, storage, or temperature changes. Priming pushes a small amount of medication out to displace any air.
- Confirms the pen is working. A drop of medication at the needle tip is the visual confirmation that the plunger, the dial, and the cartridge are all functional and connected. Without that confirmation, you cannot know if your dose actually delivered.
The priming dose is small (a fraction of a microliter, much less than a single therapeutic dose). It does not affect your weekly dose. The pen contains enough overage to allow for one or two priming attempts plus the full prescribed doses.
The official Wegovy priming protocol
Per the Novo Nordisk Wegovy Instructions for Use (the document that ships in every box and is also available on wegovy.com), the priming procedure is:
Before each first use of a new pen:
- Wash hands.
- Inspect the pen. The medication should be clear and colorless. If cloudy, discolored, or contains particles, do not use; contact the pharmacy.
- Remove the paper tab from a new needle.
- Push the needle straight onto the pen until you hear or feel a click.
- Remove the outer needle cap and save it.
- Remove the inner needle cap and discard it.
- Hold the pen with the needle pointing up.
- Turn the dose selector clockwise until the flow check symbol (a small dotted line or arrow icon) is visible in the dose window.
- Press and hold the dose button. The dose counter returns to 0.
- Look at the needle tip for a drop of medication.
If a drop appears: the pen is primed correctly. Proceed with your dose.
If no drop appears: repeat steps 8 to 10 with a new needle. You may prime up to 3 total times (one initial plus two retries).
If no drop appears after 3 attempts: do not use the pen. Contact your pharmacy or prescriber for a replacement.
When to prime twice (and what counts as a failed prime)
A "failed prime" means the dose button has been depressed all the way to the 0 mark on the dose counter, but no drop of medication is visible at the needle tip.
Things that are not a failed prime:
- A very small drop that is hard to see (try better lighting)
- A drop that forms but doesn't fall (the surface tension keeps it on the needle, which is fine)
- A small amount of liquid running down the needle shaft (counts as a successful prime)
- A drop that appears slowly over 5 to 10 seconds (still successful)
Things that are a failed prime:
- No visible liquid at all after the dose counter returns to 0
- The dose counter doesn't move when you press the button
- The dose dial gets stuck and won't turn
If the first prime fails, repeat with a fresh needle. Don't reuse the first needle. The needle change is part of the troubleshooting.
If the second prime also fails, repeat once more with another fresh needle. After three failed primes, the pen is defective and should be replaced. Most pharmacies will replace a defective pen if you contact them within a reasonable window (usually 7 days).
Common reasons a pen does not prime
A few specific issues account for most priming failures.
Air bubble blocking the cartridge. The most common cause. A pen that was shipped or stored in an unusual position can develop a large air pocket at the bottom of the cartridge. Tapping the pen gently against your palm with the needle pointing up can move the air bubble to the top, after which priming works.
Bent or clogged needle. If the needle is slightly bent during attachment, the channel can be obstructed. Always use a fresh needle for the second prime attempt. If the second needle also doesn't deliver, the issue is the pen, not the needle.
Cartridge below operating temperature. A pen pulled directly from refrigeration is too cold for the medication to flow easily. Let the pen warm to room temperature for 15 to 30 minutes before priming. The Wegovy instructions allow injection at room temperature, and warmer medication primes more reliably.
Pen has been frozen. Wegovy is destroyed by freezing. A frozen pen may look normal but won't dispense medication, or will dispense unevenly. If your pen was exposed to freezing temperatures (in shipping, in a refrigerator that ran cold, in checked luggage), discard and replace.
Pen damaged by impact. If the pen was dropped on a hard surface, internal mechanisms can be misaligned. Visible cracks or a dial that doesn't turn smoothly are signs of damage.
Empty pen. If you've already used all 4 doses and the pen is empty, the dose counter will not advance past 0. This is the pen telling you it's spent. Discard and start a new pen.
The "I primed and lost a dose" question
Patients sometimes worry that priming uses up part of their weekly dose. It does not.
Wegovy pens contain overage specifically to allow for priming. The pre-filled volume is calibrated so that the 4 weekly doses (or 1 dose, depending on which strength of pen) can all be delivered after one prime per pen, with a small buffer for one or two retries.
If you primed multiple times during the troubleshooting process, the pen still contains your full dose. The mechanism that dials up the dose for injection is independent of the priming mechanism, and the cartridge has the volume to support both.
The exception: a defective pen that delivers less than its labeled dose due to mechanical malfunction. If you suspect this happened (visibly less liquid in the cartridge than expected, dose dial behavior that seemed wrong), contact the pharmacy. They can verify and replace.
Differences between Wegovy and Ozempic priming
Wegovy and Ozempic are both made by Novo Nordisk and use similar pen platforms, but the priming protocols differ slightly.
| Feature | Wegovy pen | Ozempic pen |
|---|---|---|
| Doses per pen | 1 (single-use) for each strength | 4 (multi-dose) |
| Priming required | Before each use of new pen | Once at first use of new pen, then dose-by-dose checks |
| Flow check symbol | Yes, on each strength | Yes |
| Maximum prime attempts | 3 | 3 |
| Dose dial increments | Fixed dose (cannot be adjusted) | Variable (0.25, 0.5, 1, 2 mg available) |
The single-use vs multi-dose difference is the biggest practical distinction. A Wegovy pen is single-strength and single-use. You prime, dose, dispose. An Ozempic pen is reused for 4 weekly injections and has a different dial mechanism.
For more on Ozempic pen mechanics, see our guide on Ozempic pen clicks and dose verification.
Storage that affects pen priming
Storage conditions affect whether your pen will prime successfully. The Wegovy storage rules per the prescribing information:
Before first use:
- Store in the refrigerator at 36 to 46 F.
- Do not freeze. A frozen pen is destroyed.
- Keep the pen cap on. Protect from light.
After first use:
- Can stay at room temperature (up to 86 F) for up to 28 days.
- Or stay refrigerated (36 to 46 F) for up to 28 days.
- Discard 28 days after first use even if medication remains.
During travel:
- Insulated cooler with cold packs (not direct contact with frozen ice).
- Keep in carry-on baggage, not checked. Cargo holds can drop below freezing.
- TSA allows medication and ice packs through screening with documentation.
Common storage problems that cause priming failure:
- Pen left in a hot car (above 86 F). The medication degrades.
- Pen in a refrigerator that runs too cold (below 36 F can produce freezing).
- Pen near the back wall of the fridge where freezing is more likely.
- Pen exposed to direct sunlight on a windowsill.
If you have any doubt about storage, discard and request a replacement. The cost of one replacement pen is much lower than the cost of using a degraded pen.
Step-by-step: priming a brand-new Wegovy pen correctly
For first-time users, the full sequence:
1. Take the pen out of the refrigerator. Let it warm at room temperature for 15 to 30 minutes. Cold medication is more viscous and primes less reliably.
2. Wash your hands with soap and water.
3. Inspect the pen.
- Medication should be clear and colorless.
- No particles, no cloudiness, no discoloration.
- Pen body should be intact, no cracks.
- Dose counter should read 0.
4. Attach a new needle.
- Peel the paper tab from a new needle (NovoFine or NovoTwist work).
- Push the needle straight onto the pen.
- Screw or click into place per the needle type.
- Remove the outer needle cap (save for disposal).
- Remove the inner needle cap (discard).
5. Set the flow check symbol.
- Hold the pen with needle pointing up.
- Turn the dose selector clockwise until the flow check symbol shows in the dose window.
6. Press and hold the dose button.
- Press all the way down.
- The dose counter should return to 0.
- Watch for a drop at the needle tip.
7. Confirm the prime.
- A drop of medication at the needle tip = success.
- Proceed to your dose.
8. If no drop, repeat with a new needle. Up to 3 total attempts.
9. After successful priming:
- Set your prescribed dose using the dial.
- Inject into abdomen, thigh, or upper arm (rotate sites).
- Insert needle at 90 degrees.
- Hold the button down for at least 6 seconds after the dose counter returns to 0.
- Withdraw needle.
- Replace outer needle cap, remove needle, dispose in sharps container.
10. Storage of the partially used pen (Wegovy is single-use, so this only applies to Ozempic): replace pen cap, store at room temp or refrigerated, discard after 28 days from first use.
Troubleshooting checklist
If priming fails:
| Symptom | Likely cause | Fix |
|---|---|---|
| No drop after pressing button | Air bubble or cold medication | Tap pen with needle up, let warm to room temp, retry with new needle |
| Dose dial won't turn | Pen damaged or empty | Check for visible damage; if damaged, replace |
| Dose button won't press | Mechanical issue | Try a new needle; if still fails, replace pen |
| Drop forms slowly | Normal for cold medication | Let warm to room temp |
| Liquid runs down needle shaft | Successful prime | Proceed with dose |
| Pen makes a clicking sound but no drop | Air-only displacement | Retry with new needle in correct position |
| Visible particles in cartridge | Medication contamination | Do not use; replace pen |
| Pen feels lighter than usual | Possibly empty | Check dose counter; if pen is at end of life, replace |
FAQ
Can I prime my Wegovy pen twice?
Yes. Novo Nordisk allows up to 3 total priming attempts per new pen if a drop doesn't appear on the first try. Use a fresh needle each time.
What if my Wegovy pen never primes?
Discard after 3 failed attempts. Contact your pharmacy for a replacement. Defective pens are a known but uncommon issue and pharmacies typically replace them.
Will priming use up part of my dose?
No. Each pen contains overage specifically for priming. The 4 weekly doses (or single dose, depending on strength) are still available after priming.
Why doesn't my new Wegovy pen produce a drop?
Most often: cold medication, air bubble, or a faulty needle. Let the pen warm to room temp, tap gently, and try a new needle. If that doesn't work, the pen may be defective.
Can I prime my pen the night before injection day?
No. The pen is primed at the time of injection. Priming the night before exposes the needle to air, possible contamination, and possible medication evaporation.
Do I prime every week with the same pen?
Wegovy pens are single-use, so each new pen is primed once before its single dose. Ozempic pens are multi-dose and primed once at first use, with subsequent doses requiring only a flow-check at the start of each new pen, not before each dose.
Can I reuse a needle for the second prime attempt?
No. Always use a fresh needle for each prime attempt. The needle is part of the troubleshooting; reusing it doesn't isolate whether the pen or the needle is the issue.
What does the flow check symbol look like?
A small dashed line or stylized arrow icon on the dose dial, separate from the numeric dose markings. The exact icon varies slightly by pen strength but is consistent within a given Wegovy pen design.
Can I see the medication move through the pen during priming?
A successful prime produces a visible drop at the needle tip. You won't see medication move through the cartridge or the dial, since those are internal.
What if I forgot to prime my brand-new pen and just injected?
The dose may have been delivered short or with air. The medication likely worked, since most pens have already been pressurized at the factory and the first dose often delivers normally even without explicit priming. But you can't be certain. Tell your prescriber, watch for any unusual symptoms, and prime correctly going forward.
Does compounded semaglutide require priming the same way?
Compounded semaglutide is typically dispensed in a vial with separate syringes, not in a pen. Drawing up the dose from a vial uses different technique (no priming required, but air-bubble removal from the syringe is the equivalent step). Compounded semaglutide is not FDA-approved and is not interchangeable with brand-name Wegovy.
What if my Wegovy pen primes but the dose dial won't move past a certain number?
The pen may be defective or near end-of-life (which only matters for Ozempic, since Wegovy is single-use). Contact your prescriber or pharmacy.
