What AOD-9604 dose has been tested in human obesity research?
The best-documented large human obesity trial tested oral AOD-9604 at 0.25 mg, 0.5 mg, or 1 mg once daily for 24 weeks, alongside diet and exercise support. It did not test the 250 to 500 microgram subcutaneous schedule, fasting timing, injection technique, reconstitution recipe, or cycling advice previously presented on this page.
What did the 24-week trial find?
The FDA Pharmacy Compounding Advisory Committee briefing describes the OPTIONS study as a randomized, double-blind, placebo-controlled study in adults with obesity. Among 502 randomized participants, oral AOD-9604 at 0.25, 0.5, or 1 mg daily did not produce a statistically significant weight-loss difference from placebo at the reported endpoints. This finding applies to that oral study design and does not answer every possible research question about AOD-9604.
| Study detail | What was tested or reported |
|---|---|
| Route | Oral tablets, not subcutaneous injection |
| Daily study doses | 0.25 mg, 0.5 mg, or 1 mg |
| Duration | 24 weeks, with diet and exercise support |
| Participants | 502 randomized adults with obesity |
| Weight-loss result | No statistically significant difference from placebo reported by FDA |
What this study does not establish
An oral trial cannot be converted into a subcutaneous dose by matching the number of milligrams. The study also does not establish an empty-stomach rule, morning or bedtime timing, split dosing, an 8-to-12-week cycle, time off between cycles, dose adjustment by body size, combination protocols, or long-term use outcomes. The injection schedules and preparation directions previously shown here are not supported by this trial and have been removed.
How to read the safety summary
Stier, Vos, and Kenley summarized six randomized, double-blind, placebo-controlled AOD-9604 studies, with a focus on selected safety measures including IGF-1, glucose tolerance, and detected antibodies. The paper is a safety summary, not evidence that the subcutaneous dose ranges or cycling protocols formerly listed here were tested. Its abstract reports no treatment-related withdrawals or serious adverse events in the summarized studies; that statement should be read within the studies and follow-up included in the review.
